【摘要】:Vegetativebiofilmbacteriaarephenotypicallydifferentfromsuspendedcellsofthesamegenotype.Biofilmgrowthreactorsareengineeredtoproducebiofilmswithspecificcharacteristics.Alteringeithertheengineeredsystemoroperatingconditionswillmodifythosecharacteristics.Thepurposeofthistestmethodistodirectauserinhowtogrow,sampleandanalyzeaPseudomonasaeruginosabiofilmunderlowfluidshearandclosetotheair/liquidinterfaceusingthedripflowreactor.ThePseudomonasaeruginosabiofilmthatgrowshasasmoothappearanceandislooselyattached.Microscopically,thebiofilmissheet-likewithfewarchitecturaldetails.Thislaboratorybiofilmcouldrepresentthosefoundonproducesprayers,onfoodprocessingconveyorbelts,oncatheters,inlungswithcysticfibrosisandoralbiofilms,forexample.Thebiofilmgeneratedinthedripflowreactorisalsosuitableforefficacytesting.Afterthe54hgrowthphaseiscomplete,theusermayaddthetreatmentinsituorharvestthecouponsandtreatthemindividually.ResearchhasshownthatP.aeruginosabiofilmsgrowninthedripflowreactorwerelessresistanttodisinfectionthanbiofilmsgrownunderhighshearconditions.1.1ThistestmethodspecifiestheoperationalparametersrequiredtogrowarepeatablePseudomonasaeruginosabiofilmclosetotheair/liquidinterfaceinareactorwithacontinuousflowofnutrientsunderlowfluidshearconditions.Theresultingbiofilmisrepresentativeofgeneralizedsituationswherebiofilmexistsattheair/liquidinterfaceunderlowfluidshearratherthanrepresentativeofoneparticularenvironment.1.2Thistestmethodusesthedripflowbiofilmreactor.Thedripflowreactor(DFR)isaplugflowreactorwithlaminarflowresultinginlowfluidshear.Thereactorisversatileandmayalsobeusedforgrowingand/orcharacterizingdifferentspeciesofbiofilms.1.3Thistestmethoddescribeshowtosampleandanalyzebiofilmforviablecells.Biofilmpopulationdensityisrecordedaslogcolonyformingunitspersurfacearea.1.4Basicmicrobiologytrainingisrequiredtoperformthistestmethod.1.5ThevaluesstatedinSIunitsaretoberegardedasstandard.Nootherunitsofmeasurementareincludedinthisstandard.1.6Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.

【摘要】:Theconstanttensileloadjointtest(CTLJT)isdesignedtodemonstratethatajointinaplasticpipingsystemisresistanttotheeffectsoflong-termcreep.Thejointissubjectedtoaninternalpressureatleastequaltoitsoperatingpressureandasustainedaxialtensileloadforaspecifiedtimeperiod,usually1000h.Thejointshallnotleak,normaythepipecompletelypulloutforthetestduration.Thetotalaxialstressissetbythereferencingdocument.SometypicalconditionsfortestingofjointsonpolyethylenepipearedescribedinAppendixX1.Thistestisusuallyperformedat73?F(22.8?C).TheCTLJTwasdevelopedtodemonstratethelong-termresistancetopulloutofmechanicaljointsonpolyethylenegaspipe.TheCTLJThasalsobeensuccessfullyappliedtotheevaluationofothercomponentsofplasticpipingsystems.TheseapplicationsarediscussedinAppendixX1.Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
